چکیده

The detection of a supranormal quantity of immunoglobulin G (lgG) in cerebrospinal fluid is valuable in diagnosis of multiple sclerosis. In a previously published study of over 300 patients, including 46 patients with a clinical diagnosis of multiple sclerosis, 74% of the patients were found to have an abnormally high lgG value, expressed as a ratio of lgG/albumin, by the reliable method of electroimmunodiffusion. In the presont study the recently introduced method of radial immunodiffusion was compared with established electroimmunodiffusion technique in divided samples of cerebrospinal fluid from 49 patients with a wide range of lgG/albumin values. The correlation of the results obtained by these two methods was very good (r = 0.945), but the variance with radial immunodiffusion (±20 % ) is greater than with electroimmunodiffusion (± 15 % ). Two samples that exhibited increased values by the electroimmunodiffusion method were normal with the other technique. We condude that electroimmunodiffusion remains a slightly more accurate and sensitive technique, although radial immunodiffusion is simpler and requires less technician time.

Methodologic differences in values for M-proteins in serum, as measured by three techniques.

Accurate and reproducible measurement of M-proteins is essential for managing patients with monocional gammopathies, but serum protein electrophoresis, radial immunodiffusion, and electroimmunodiffusion yield comparatively divergent results. We have studied these differences and their causes. Sera from cases of IgG-monoclonal gammopathy, IgA-monoclonal gammopathy, and IgM-monoclonal gammopathy ...

Comparison of protein values in cerebrospinal fluid by nephelometry and radial immunodiffusion.

Various parameters of three radial immunodiffusion systems, a kinetic nephelometer and two laser nephelometers were compared in a measurement of IgG and albumin in cerebrospinal fluid. Greater precision for IgG and albumin determinations was found with two RID systems. Correlation coefficients between methods ranged from 0.71 to 0.94. The generally poor correlations of methods, combined with th...

Rapid method for protein quantitation by electroimmunodiffusion.

In the electroimmunodiffusion method described, a Mylar-backed cellulose acetate membrane plate is used as a support medium. Results are obtained faster, less preparation time is required, and the method is less expensive because less antisera is needed. Results with the method are reproducible (CV, 5%), correlate well with results by the single radial immunodiffusion method, and sensitivity is...

Use of laser nephelometry in the measurement of serum proteins.

Measurement of immunoglobulins (Ig) and of complement component C3 in human serum by automated and manual nephelometric techniques is tedious, and the effective linear range is too narrow. We describe a laser nephelometer/reagent system for measuring serum proteins by the use of forward light scatter (which enhances the ratio of reaction/blank), electronic blank subtraction, laser light (632 nm...

Comparison of radial immunodiffusion and laser nephelometry for quantitating some serum proteins.

After quantitating immunoglobulins G, A, and M and complement C3c and C4 in serum by using a laser nephelometer coupled with a data processor, I compared these results with values obtained by an early-readout radial immunodiffusion method. Day-to-day precision was better for nephelometry than for radial immunodiffusion for all proteins analyzed. The average coefficient of variation was 6.0% for...
